THE Spireites' interest in the Carling Cup came to an end despite a gallant fight to overcome opponents two divisions higher.
Scunthorpe scored early through Matt Sparrow and doubled their lead when the predatory Paul Hayes scored from close range early in the second half.
Sub Darren Currie came on and almost immediately scored from the spot after Drew Talbot was fouled but despite plenty of endeavour, Chesterfield were unable to find an equaliser.
John Sheridan made six changes to the side that lost at Torquay.
In came Tommy Lee, Martin Gritton, Danny Hall, Dan Gray plus Jordan Bowery - starting for the first time ever - and Paul Harsley, in the first eleven for the first time since Boxing Day against one of his previous clubs.
Onto the bench went Mark Crossley, Phil Picken, Rob Page, Derek Niven and Currie while Jamie Lowry missed out completely.
The Championship side took the lead after just 12 minutes following an all-action start by both sides.
Gary Hooper broke down the right and delivered an inch-perfect cross for Sparrow to roll home, unmarked at the far post just seconds after Marcus Williams had been forced to head a Donal McDermott right wing corner from off the line.
Gray, at right back, was having a difficult time against Martyn Woolford and was soon replaced by Picken.
Mark Allott had a shot blocked before Lee was forced into serious action, diving full stretch to his right to palm over a fierce Hooper shot.
Michael O'Connor gave a warning about the quality of his free kicks as he drifted one straight into the net, but the effort was ruled out for a foul, but his next similar effort forced a reflex save from Lee only for Hayes to volley home from close range.
Hayes saw one shot deflected inches wide and another hit the goal frame ahead of Kevin Austin somehow heading a Woolford cross from right underneath the bar and out for a corner.
With the Iron in charge, Talbot darted into the box, checked and was upended by Rob Jones.
Currie, on the park for just five minutes, stepped up to score the penalty to give his side their first competitive goal in over 11 hours of play.
